Understanding how DailyDIM+™ works requires understanding how estrogen is actually metabolized — and where things can go wrong.

Phase I — Estrogen Conversion (Hydroxylation) In phase I liver detoxification, estrogen is hydroxylated into one of three metabolite pathways: 2-hydroxyestrone (2-OHE1), 4-hydroxyestrone (4-OHE1), or 16α-hydroxyestrone (16α-OHE1). The 2-OH pathway is generally considered the favorable route — producing metabolites with weaker estrogenic activity. The 4-OH and 16α-OH pathways produce more potent, potentially problematic metabolites. Supporting the shift toward 2-hydroxylation is the primary goal of DIM supplementation.*

Phase II — Glucuronidation and Conjugation Once hydroxylated, estrogen metabolites undergo phase II detoxification — primarily glucuronidation, where the liver attaches glucuronic acid to the metabolite, rendering it water-soluble and prepared for excretion via bile or urine. This is where Calcium-D-Glucarate and sulforaphane play their critical roles, supporting the enzymatic activity that completes this preparation step.*

Phase III — Elimination (Preventing Reabsorption) This is the step most hormone supplement formulas miss entirely. Even after the liver has successfully glucuronidated estrogen metabolites, they still travel through the intestines on their way out. In the gut, an enzyme called beta-glucuronidase — produced by certain gut bacteria — can reverse the glucuronidation process, cleaving the glucuronic acid bond and freeing the estrogen metabolite to be reabsorbed into circulation through enterohepatic recirculation.*

Calcium-D-Glucarate inhibits beta-glucuronidase activity, ensuring glucuronidated estrogens remain bound and are eliminated rather than recycled. Without this step, the liver's phase II work can be undone in the gut.*

THE FIVE-INGREDIENT FORMULA

DIM (Diindolylmethane)

Dose: 300 mg Role: Phase I Estrogen Metabolism Support

DIM is a naturally occurring phytonutrient derived from indole-3-carbinol (I3C), produced during digestion of cruciferous vegetables including broccoli, cauliflower, kale, and Brussels sprouts. As an active metabolite of I3C, DIM is more bioavailable and more targeted in its activity than its precursor.

DIM's primary mechanism is support of cytochrome P450 enzyme activity in the liver — specifically CYP1A1 and CYP1A2 — which drive the 2-hydroxylation of estrogen toward the 2-OH pathway and away from the 4-OH and 16α-OH pathways.* By promoting a more favorable 2-OH:16α-OH ratio in urinary estrogen metabolites, DIM supports healthier estrogen signaling balance.*

DIM also supports healthy androgen-estrogen balance in both men and women by modulating aromatase activity and SHBG levels — making it relevant not only for women navigating hormonal fluctuation but also for men concerned with estrogen balance in the context of healthy testosterone metabolism.*

Calcium-D-Glucarate

Dose: 1,000 mg Role: Phase II Glucuronidation Support & Beta-Glucuronidase Inhibition

Calcium-D-Glucarate is the calcium salt of D-glucaric acid — found naturally in cruciferous vegetables, oranges, and apples — metabolized in the body to D-glucaro-1,4-lactone, one of the most potent natural inhibitors of beta-glucuronidase.*

Its primary mechanism works at the gut level: by inhibiting beta-glucuronidase, Calcium-D-Glucarate prevents the deconjugation of glucuronidated estrogens and toxins in the intestine — the critical final step that ensures that what the liver has processed for elimination is actually eliminated rather than reabsorbed.*

This mechanism is particularly important because enterohepatic recirculation of estrogen can meaningfully increase effective estrogen exposure even when phase I metabolism is well-supported — a gap that DIM alone cannot address.*

Activated BroccoRaphanin™

Dose: 150 mg (Broccoli Seed Concentrate + Myrosinase Enzyme) Role: Nrf2-Mediated Phase II Detoxification & Cellular Antioxidant Defense

Activated BroccoRaphanin™ delivers sulforaphane glucosinolate (SGS) from broccoli seed concentrate, standardized for consistent glucoraphanin content, paired with myrosinase enzyme — the specific enzyme required to convert glucoraphanin into bioactive sulforaphane.* This pairing is critical: cooking and processing destroy myrosinase activity in most broccoli-based foods and supplements, meaning glucoraphanin consumed without myrosinase produces little active sulforaphane. Activated BroccoRaphanin™ preserves and delivers both.*

Sulforaphane is widely recognized as one of the most potent known activators of the Nrf2 (Nuclear factor erythroid 2-related factor 2) transcription pathway — the body's master switch for phase II detoxification enzyme induction and antioxidant defense.* When sulforaphane modifies Keap1 — its molecular target — Nrf2 translocates to the nucleus, where it binds the Antioxidant Response Element (ARE) and upregulates over 200 cytoprotective genes.* These include:
